As long as the trans isn't shifting in and out constantly, you shouldn't have any problems towing in O/D.

I just moved from Homestead Fl to Gainesville Fl using a rented trailer. I took me three trips with the trailer to make the move. The '09 has a tow/haul mode that I don't think applies to your '01 but the principal is the same. Over basicly flat land the only difference you'll see with O/D "off" is lower mpg. However, as Remerson said, if your hunting for a gear a lot you are best to drop out of o/d to save wear and tear on the tranny.

I would say as long as your up to speed and don't have a probem maintaining it ,you should be ok to activate O/D . Once you get into some hilly areas shut it off .

I have a 03 3500 and never take mine out of OD and I pull a 6000 pound camper. If I hit a big hill I just ease off the throttle a bit and let her do her thing.

I never take the OD off on my Ram 1500, and I never use the manual shift on my Ram 3500. I just stick it into drive and haul away. Never had tranny issues. Got the 545RFE on the Ram 1500 and the 68RFE in the Ram 3500. But like everyone else said, if you feel that it's shifting in and out of gears frequently then switch the O/D off.

I haul both a 9600lb trailer and a 12000lb trailer with my '09 superduty, but the same theory still applies. When I'm at highway speed, i'll leave the O/D on, as well as just regular starting out. With the 12000lb trailer I'll take it off to get going from a stop, but then i'll turn it back on. I only use it when I'm going through steep terrain or when I'm off road towing to a lease site.
